New Retinoids for Dermatologic Diseases: Uses and Toxicity

The synthetic retinoids—isotretinoin, etretinate, acitretin, arotinoid ethyl ester—are frequently highly effective against dermatologic conditions including acne, disorders of cornification, and severe psoriasis. Careful patient selection and dosing are required, and there are significant toxicity problems. Systemic administration of retinoids is frequently associated with mucocutaneous side effects, liver toxicity, and abnormalities of the serum lipid profile. Currently, the principal factors limiting retinoid use are their teratogenicity and chronic bone toxicity.
